I just started playing the game, so I only finished Tokyo so far. I have only unlocked Black Cat, Doc Oc and the Green Goblin, plus the two I started out with. So far I like Black Cat the best. Oh and I have a little card from Gamestop that came with the game that aparently allows me to open up and use Venom with some button code input. Is that the only way to open him? Or can you open him during the course of the actual career mode of the game? I ask because i don't want to use cheats.

you can unlock venom. but you have to finish tokyo, tangaroa island, egypt, and then beat him in transylvania.
