To unlock Challenge Mode, you must defeat the 2nd Bowser. Challenge Mode will be available on the Title Screen and contains a series of extra minigames. There are 5 Challenge Marbles for that mode.

Unlock 1st Marble: Defeat the 2nd Bowser
Unlock 2nd Marble: Complete the Egg Gallery
Unlock 3rd Marble: Complete the Egg Gallery
Unlock 4th Marble: Complete the Egg Gallery
Unlock 5th Marble: Get EVERY Gold Medal.
